Light intensity has an important influence on the proliferation of cultured cells and the differentiation of organs. According to the current research, light intensity has an obvious influence on the initial division of plant cells. Generally speaking, light intensity is strong, and the seedlings grow strongly, while when light intensity is weak, the seedlings tend to grow slender. There is no doubt that the light intensity tissue require is not as strong as that of robust plant. 

Light quality also plays an important role in the process of callus induction, proliferation of cultured tissue, and organ differentiation. For example, when Lily tissues were cultured under red light. Callus generates after 8 weeks. However, under blue light, callus appears after just several weeks.  If the culture calamus tissues in blue light. After 15 days, you will find seeds in vigorous growth; while under white light, the seedlings are slender.

Study shows that monochromatic light has an inhibitory effect on chlorophyll synthesis. The synthesis of chlorophyll needs to be done under the condition of compound light.

Tube tissue cultivation should choose a suitable photoperiod to conduct tissue culture. The most commonly used photoperiod is 16 hours in light and 8 hours in darkness. Research shows that plant organs of short-day plants are more likely to differentiate in short sunlight lighting conditions. While under long sunlight, plant organs of short-day plants are more inclined to produce callus. Some plant callus cultured in dark condition is better than under the condition of light, such as the callus of safflower and blueberry. With the exception of some special plants, most general tissue cultures require a certain amount of light.
